This weekend most of you will be at home, catching up with family, sharing a meal and giving thanks. The only risk involved in this process is the high chance of bickering. When the festivities are over though, a lot of you will make a long journey home by car. That?s something that does require a bit of risk management.

The long drive home actually makes a great point about the risk management process. Long drives are one of those things that get a lot of hair-brained risk management. There is a certain amount of conventional wisdom that gets passed around about long drives, very little of it is useful and most of it is dangerous.

Conventional Wisdom and Risk

The big risk associated with a long drive is the risk of falling asleep at the wheel. You?ve had a celebratory weekend. You?ve had drinks, late nights and lots of food, when you get into that car you?re likely to be tired. So you apply the traditional risk management process to the drive. You do one or all of the following:

-Drive with the windows down

-Play loud music

-Drink lots of coffee

Each of these is supposed to keep you awake. They might stave off sleep for a little while, but each one is an awful idea. The loud music makes you less aware of what?s happening around you. The window just makes you cold and distracted by the rush of air and coffee used as a stimulant is the definition of a temporary measure. Stimulants work for a while but the energy is artificial, you know what happens when that energy dissipates? You crash, figuratively and literally.

The Risk Management Process

A proper risk management process ignores these temporary or flawed methods. A proper process involves the methodical assessment and management of risk. It involves preventing risk through statistically proven methods. True risk management involves the assessment of everything from the risks to the risk management process itself.

The lesson is simple, don?t rely on conventional wisdom or that free advice you got from a buddy to manage risk. In your personal life, as well as in business, you should always talk to an expert about risk management.

The cerebral cortex of the human brain has been called "the crowning achievement of evolution." Ironically, it is so complex that even our greatest minds and most sophisticated science are only now beginning to understand how it organizes itself in early development, and how its many cell types function together as circuits.

A major step toward this great goal in neuroscience has been taken by a team led by Professor Z. Josh Huang, Ph.D., at Cold Spring Harbor Laboratory (CSHL). Today they publish research for the first time revealing the birth timing and embryonic origin of a critical class of inhibitory brain cells called chandelier cells, and tracing the specific paths they take during early development into the cerebral cortex of the mouse brain.

These temporal and spatial sequences are regarded by Huang as genetically programmed aspects of brain development, accounting for aspects of the brain that are likely identical in every member of a given species including humans. Exceptions to these stereotypical patterns include irregularities caused by gene mutations or protein malfunctions, both of which are now being identified in people with developmental disorders and neuropsychiatric illnesses.

Chandelier cells were first noticed only 40 years ago, and in the intervening years frustratingly little has been learned about them, beyond the fact that they "hang" individually among great crowds of excitatory cells in the cortex called pyramidal neurons, and that their relatively short branches make contact with these excitatory cells. Indeed, a single chandelier cell connects, or "synapses," with as many as 500 pyramidal neurons. Noting this, the great biologist Francis Crick decades ago speculated that chandelier cells exerted some kind of "veto" power over the messages being exchanged by the much more numerous excitatory cells in their vicinity.

Born in a previously undiscovered 'country'

After three years of painstaking work that has involved using new technologies to identify and trace neural cell progenitors in ways not previously possible, and to track them as they migrate to positions in the maturing cortex, Huang and colleagues, including Dr. Hiroki Taniguchi now at the Max Planck Florida Institute, have demonstrated that chandelier cells are born in a previously unrecognized portion of the embryonic brain, which they have named the VGZ (ventral germinal zone).

Huang, who has been on a decade-long quest to develop means of learning much more about the cortex's inhibitory cells (sometimes called "interneurons"), points out that while they are far less numerous than the excitatory pyramidal cells all around them, cells including chandelier cells that inhibit or modulate excitatory-cell messages play an indispensable role in balancing message flow and ultimately in determining the functional organization of excitatory neurons into meaningful groups.

This is all the more intriguing in the case of chandelier cells, Huang explains, because of their distinctive anatomy: one cell that can regulate the messages of 500 others in its vicinity is one that we need to know about if we want to understand how brain circuits work. Unlike other inhibitory cells, chandelier cells are known to connect with excitatory cells at one particular anatomical location, of great significance: a place called the axon initial segment (AIS) ? the spot where a "broadcasting" pyramidal cell generates its transmittable message. To be able to interdict 500 "broadcasters" at this point renders a single chandelier cell a very important player in message propagation and coordination within its locality.

Because of the strategic importance of such cells throughout the cortex, it has been a source of frustration to neuroscientists that they (and other inhibitory cells) have been difficult to classify. Huang has pursued a strategy of following them from their places of birth in the emerging cortex.

Many inhibitory cells come from a large incubator area called the MGE (medial ganglionic eminence); until now, it was not known that most chandelier cells are not born there, and indeed do not emerge until after the MGE has disappeared. Only at this point does the much smaller VGZ form, providing a place where neural precursor cells specifically give rise to chandelier cells.

The team learned that manufacture of a protein encoded by a gene called Nkx2.1 is among the signals marking the birth of a chandelier cell. The gene's action, they found, is also necessary to make the cells. Nkx2.1is a transcription factor, whose expression has previously been linked to the birth of other inhibitory neuronal types. Huang's team observes that it is the timing of Nkx2.1's expression in certain precursors -- following disappearance of the MGE and appearance of the VGZ -- that enabled them to track the birth, specifically, of chandelier cells.

Highly specific migration route and cortical destinations

"In addition to being surprised to discover that chandelier cells are born 'late'?after other inhibitory cells ? in a part of the cortex we didn't know about," says Huang, "our second surprise is that once born, these cells take a very stereotyped route into the cortex and assume very specific positions, in three cortical layers." (Layers 2, 5 and 6). "This leads us to postulate that other specific cortical cell types also have specific migration routes in development."

As Huang points out, his team's new discoveries about chandelier cells have implications for disease research, since it is known that the number and connective density of chandelier cells is diminished in schizophrenia. Associations of the same type have recently been made in epilepsy.

"To know the identity of a cell type in the cortex is in effect to know the intrinsic program that distinguishes it from other cell types. In the broadest terms, we are learning about those aspects of the brain development that make us human. 'Nurture,' or experience, also has a very important role in brain development. Our work helps clarify the 'nature' part of the nature/nurture mystery that has always fascinated us," Huang says.

Stocks soar on Black Friday; tech leads the way

The stock market enjoyed some Black Friday cheer, rising sharply as shoppers braved the annual post-Thanksgiving rush. Major stock indexes closed one of their best weeks of the year.

Traders were encouraged by positive economic news from Germany and China, two engines of global growth. Technology stocks soared after a few weeks of selling. And early reports from retailers suggested strong consumer spending.

"Foot traffic appears heavier than we've seen in recent years, there are a lot of positive statements out of the companies themselves, and momentum appears to be strong," said JJ Kinahan, chief derivatives strategist at the brokerage TD Ameritrade.

Many stores opened earlier than ever this year, Kinahan said, allowing for earlier informal reports about their performance.

Technology stocks soared, lifting the Nasdaq composite index by more than 1 percent. Dell, chipmaker AMD and Hewlett-Packard were the top three gainers in the Standard & Poor's 500. Technology rose the most among the index's 10 industry groups.

The stocks were bouncing back after confidence in tech stocks declined broadly, Kinahan said. AMD dropped sharply in recent weeks as investors fretted about its solvency. HP plunged 12 percent on Tuesday after executives said that a company HP bought for $10 billion last year lied about its finances.

The Nasdaq ended up 40.30 points, or 1.4 percent, at 2,966.85. The Dow Jones industrial average gained 172.79, or 1.4 percent, to 13,009.69 ? the first time since election day that the Dow closed above 13,000.

The S&P 500 added 18.12, or 1.3 percent, to 1409.15. The rally gave the S&P 500 its biggest weekly point gain since last December ? 49 points, or 3.6 percent. The Dow gained 3.4 percent and the Nasdaq almost 4 percent for the week.

The market closed early, at 1 p.m. EST.

Stocks started strong after news that German business confidence rose unexpectedly in November after six straight declines. The gain in a closely watched index published by Munich's Ifo institute raised hopes that Europe's largest economy can continue to weather the continent's financial crisis.

China's manufacturing expanded for the first time in 13 months in November, the latest sign that the world's second-biggest economy is recovering from its deepest slump since the 2008 global crisis. HSBC Corp. said its monthly Purchasing Managers' Index improved to 50.4 for November. Any number above 50 indicates expansion.

The PMI measures overall manufacturing activity by surveying indicators including orders, employment and production. The result was released Thursday, when the U.S. market was closed for Thanksgiving.

Around the U.S., shoppers flocked to malls and logged on to computers to take part in the annual cheer-fueled retail rush known as Black Friday.

Target and Toys R Us welcomed buyers on Thanksgiving evening. Retailers are also trying to draw shoppers with free layaway and shipping, by matching prices of online rivals and by beefing up mobile shopping apps.

Retail is a key driver of the nation's economy. Consumer spending accounts for about 70 percent of U.S. economic activity. November and December, which can account for as much as 40 percent of a retailer's annual revenue, are crucial for stores.

The Friday after Thanksgiving is known as Black Friday because it begins the period in which many retailers turn profitable for the year. Traders will be looking for signs about how enthusiastically Americans are spending. That could reflect the momentum of the economic recovery.

Wal-Mart rose $1.31, or 1.9 percent, to $70.20. Macy's gained 72 cents, or 1.8 percent, to $41.73.

Trading volume on Wall Street was light, with many investors away for an extended weekend after Thanksgiving. The rally's strength will be tested on Monday, as many traders return to their desks and retailers begin to release hard data about their holiday sales results, Kinahan said.

"It's great when something like this happens, but on a half-day without the major players in there, it's not so surprising," he said.

European indexes added to earlier gains after Wall Street opened and closed higher. The FTSE 100 index of leading British shares rose 0.5 percent. Germany's DAX and France's CAC-40 both added 0.9 percent.

Investors were monitoring developments in Brussels, where European Union leaders are trying to agree on a $1.25 trillion long-term spending plan for the 27-nation bloc. Markets expect that another meeting will be needed for an agreement.

Among the stocks making big moves:

? MAP Pharmaceuticals spiked after the company announced that the Food and Drug Administration will review its experimental migraine drug Levadex. The stock rose $2.60, or 20.3 percent, to $15.42.

? KIT Digital Inc., a video software and technology company, lost two-thirds of its value after the company's former chief executive accused it of blaming prior management for its financial problems. Two days earlier, KIT said it would restate its financial results because of accounting errors. The stock lost $1.33, or 64.3 percent, to 74 cents.

Among tech's big gainers:

? Research in Motion Ltd. jumped on growing optimism for an earlier-than-expected launch of its delayed BlackBerry 10 smartphone. A senior executive from the Canadian company said earlier this month that Research In Motion, or RIM, will release the latest version of its smartphone "not long after" a Jan. 30 event. One analyst saw that as an indication that the products are to be unveiled in February. U.S.-traded shares of RIM rose $1.40, or 13.7 percent, to $11.66.

? Dell rose 49 cents, or 5.4 percent, to $9.55.

? HP added 50 cents, or 4.2 percent, to $12.44.

? AMD rose 8 cents, or 4.3 percent, to $1.95.

Wreck victims on family's first Thanksgiving apart

BEAUMONT, Texas (AP) ? The children of a Houston-area couple killed during Thursday's massive interstate wreck in Texas say this year was the family's first Thanksgiving apart.

Authorities are investigating the chain-reaction wreck that involved more than 100 vehicles, many of which were left twisted on top of each other. The wreck killed the couple and injured at least 80 other people during heavy fog along Interstate 10 near Beaumont.

Twelve people remain hospitalized in serious condition, the Texas Department of Public Safety said in a statement Friday.

Vincent and Debbie Leggio were on their way to Louisiana for a gambling trip when their vehicle was caught in the wreck, sons V.J. and Richard Leggio told KHOU-TV.

"My parents are wonderful loving people ? all who knew them knew that," V.J. Leggio told the station. "They will be sorely missed ? sorely missed."

He said he worked with his father at the family business, Vinco Electric in Pearland, the Houston suburb where his parents lived.

"We had a good relationship," he added. "We got to spend time together golfing and fishing, and I'm so thankful for that now."

The wreck occurred southwest of Beaumont, on Texas' Gulf Coast. The heavy fog made it hard for law enforcement to determine at first how many wrecks had occurred.

According to the Texas Department of Public Safety, a crash on the eastbound side of the highway led to other accidents in a dangerous chain reaction. There were multiple crashes on the other side of the highway as well.

Texas Department of Public Safety Sgt. Chuck Havard told KFDM-TV on Friday that investigators will work to determine whether any drivers will face charges.

Phone messages left Friday by The Associated Press for law enforcement officials weren't returned.
